Welcome. And sorry for the mistake - Vardzia is of course west of Akhalkalaki, and not east of it ;-)
Anyway - the place to visit at least once in a lifetime.

No, they are not close. Uplistsikhe is near Gori, not far from Tbilisi. While Vardzia is almost at the border with Turkey, east of Akhalkalaki. But both Uplistsikhe and Vardzia are on the bank of Mtkvari river - the same which goes through Tbilisi.
